چکیده

Abstract Background Valproic acid (VPA) is a first-line antiepileptic drug for glioblastoma (GBM) patients. There also some evidence it improves the clinical outcome in those However, exact mechanism of VPA action vague. Temozolomide (TMZ) chemotherapeutic GBM. Epigenetics offers connection between genetic and environmental factors that influence development disease. The best-characterized epigenetic mark 5-methylcytosine (m5C) DNA. aim project to show effects administration on total DNA methylation level. Material Methods Using nucleotide post-labeling method, we analyzed amount m5C GBM (T98G, U118, U138), cancer (HeLa) normal (HaCaT) cell lines treated with VPA, combination TMZ. Results We adjusted doses ones achieved central nervous system during treatment. observed dose-dependent changes neoplastic lack such effect line. alone produced clear increase scarce non-neoplastic In lines, TMZ decreased level m5C. exposition cells caused an adverse synergistic resulting demethylation. highest loss was at concentration (100 μM) (350 μM), after 3 h incubation T98G Conclusion Total glioma under treatment suggest new promote implications adjusting therapy Our results potential possible obstacles combined VPA. experiments both drugs leads hypomethylation. Therefore conclusion would be stop chemotherapy temporarily.

In Vitro Radiosensitizing Effects of Temozolomide on U87MG Cell Lines of Human Glioblastoma Multiforme

Background: Glioma is the most common primary brain tumor with poor prognosis. Temozolomide (TMZ) has been used with irradiation (IR) to treat gliomas. The aim of the present study was to evaluate the cytotoxic and radiosensitizing effect of TMZ when combined with high-dose and high-dose rate of gamma irradiation in vitro. Methods: Two ‘U87MG’ cell lines and skin fibroblast were cultured and ass...

Transcriptional regulation of E-cadherin and oncoprotein E7 by valproic acid in HPV positive cell lines

Objective(s): Valproic acid (VPA) has proven to be as one of the most promising useful drug with anticancer properties. In this study, we investigate the VPA effects on E-cadherin expression in HeLa, TC1, MKN45, and HCT116 cell lines.  This study assesses the effects of VPA on human papillomavirus E7 expression in HPV positive cell lines. Materials and Methods: Cell lines were treated by2 mmol/...

Prolonged survival with valproic acid use in the EORTC/NCIC temozolomide trial for glioblastoma.

OBJECTIVE This analysis was performed to assess whether antiepileptic drugs (AEDs) modulate the effectiveness of temozolomide radiochemotherapy in patients with newly diagnosed glioblastoma. METHODS The European Organization for Research and Treatment of Cancer (EORTC) 26981-22981/National Cancer Institute of Canada (NCIC) CE.3 clinical trial database of radiotherapy (RT) with or without temo...

Valproic acid related idiosyncratic drug induced hepatotoxicity in a glioblastoma patient treated with temozolomide.

Glioblastoma patients undergoing treatment with surgery followed by radiation and temozolomide chemotherapy often develop a state of immunosuppression and are at risk for opportunistic infections and reactivation of hepatitis and herpes viruses. We report the case of a 48-year-old glioblastoma patient who developed acute cholestatic hepatitis with hepatic failure during adjuvant treatment with ...

Data on novel DNA methylation changes induced by valproic acid in human hepatocytes

Valproic acid (VPA) is a widely prescribed antiepileptic drug in the world. Despite its pharmacological importance, it may cause liver toxicity and steatosis. However the exact mechanism of the steatosis formation is unknown. The data presented in this DIB publication is used to further investigate the VPA-induced mechanisms of steatosis by analyzing changes in patterns of methylation. Therefor...
